The best April Fools joke I heard was this:

Create an hour long or so .wav file that starts with the windows boot sound, after that, silence. Most of the .wav file should be silent. But, at random intervals, insert some sounds. Maybe whisper their name, maybe put in a fart noise, a beep, whichever. Be creative.

And when they aren't around, replace their windows boot sound with this.

Nobody suspects the spanish inq--err, boot sound.
